LEAP™ (Laser-Enabled Analysis and Processing)
LEAP™
is an automated live cell analysis and processing system for in situ laser-based cell purification and/or transfection. LEAP™ combines ultra high-speed imaging and multi-parameter visual characterization of cells with simultaneous laser-based cell manipulation. LEAP™ can image and process adherent and non-adherent cells using C-lect™ applications kits and microplates. Capabilities of LEAP™ include:

  • in situ imaging of cells using brightfield and multi-wavelength fluorescence characteristics; and
  • laser-based manipulation to perform cell purification and/or optoinjection.
The LEAP™ system offers a truly unique and powerful means to combine live cell imaging and cell manipulation in an automated mode. Since LEAP™ exploits standard 96- and 384-well microplate formats, cell processing on LEAP™ and other automated procedures, such as cell culture and liquid handling, can be easily integrated.

Key LEAP™ advantages include:

  • iterative in situ cell processing to help maximize desired cell purity and yield;
  • laser-based delivery of a wide variety of molecules into cells;
  • laser-based manipulation of a wide variety of sample sizes;
  • processing of many cell types, including  stem cells, fragile cells and primary cells; and
  • safer processing of certain biohazardous cells.

CellXpress powered by LEAP

CellXpress™ exploits high-speed imaging of living cells combined with simultaneous laser elimination of undesired cells, leaving behind cells that are efficiently secreting biopharmaceutical proteins.  The process is initiated by imaging hundreds of cells per well to quantify the level of protein being secreted by each cell. The best secreting cell is located in seconds within a pool of hundreds of candidate cells, and once located; all other undesired cells are selectively eliminated with the laser leaving behind only the best cell.  Unlike other cell purification methods, the cell of interest remains completely undisturbed during the process. This purification results in a verified clone that will generate a highly stable secreting cell line.  The imaging and purification processes can be repeated hundreds of times an hour to enable scanning through millions of cells per day, often yielding hundreds of highly-secreting clones.

Diagram and real image demonstrating detection of living cell and secreted protein. Live cells are identified in green, and associated secreted protein is visible as a red halo around each cell. Image on right was captured on the LEAP™ platform.

CellXpress™ benefits include:

  • Isolate up to ten-fold more highly secreting clones in less time
  • Less than half the time to validated clones
  • Up to 90% reduction in labor and material costs
  • Process multiple projects / cell lines in parallel
  • Process cells directly in growth medium
  • Process cells in sterile, closed plate environment
  • Higher throughput / scalable / automatable
