Marker Analysis
Analysis of transcription factors, cell signaling events, and cell surface marker expression can be accurately quantified using the Celigo. The Celigo provides a unique combination of brightfield and fluorescence imaging, image segmentation, and analysis tools for researchers to quickly evaluate routine immunocytochemical staining and to perform high-throughput screens. Assays can be multiplexed for detection and quantification of multiple markers simultaneously. While subsampling of a well can be used to correctly analyze individual cells, whole-well images are necessary to accurately analyze the marker expression of colonies, such as stem cell colonies. Marker analysis on Celigo provides confers several advantages:
- Whole-well imaging and analysis of multi-well plates (scan and analyze an entire 96-well plate in <16 min)
- Easy-to-use interface to image and analyze markers in a wide variety of cell types
- Advanced flow-like gating interface enables subpopulation analysis
- Analyze marker expression of cells and colonies
- Plate level overview of analyzed, quantitative results
Marker analysis on Celigo. Images of hiPSCs on matrigel stained with SSEA4 (top) and segmented (bottom) using the Expression Analysis application. Nuclei are segmented as the mask and SSEA4-positive nuclei are segmented as the target to determine the percentage of cells expressing SSEA4.
Analysis of nuclear marker expression on Celigo. Images of reprogrammed fibroblasts stained with KLF4 (top, bottom left) and segmented (bottom right) using the Expression Analysis application.
