Isolation of specific stem cell colonies is a key step in the generation of induced pluripotent stem (iPS) cell lines and of genetically modified stem cell lines. Typically done manually under a dissection microscope, this requires expertise and is not compatible with either automation or the use of fluorescent markers to guide colony selection. The Stem Cell Colony Purification on LEAP provides a user friendly, reproducible platform for the purification of colonies. Using high speed brightfield and/or fluorescence whole well imaging, colonies are interactively selected and purified via laser processing. The ability to isolate specific colonies in situ provides several important benefits to the stem cell user:

  • Easily monitor growth of specific colonies to guide selection of putative iPS colonies in reprogramming experiments.
  • Use fluorescent markers to identify colonies of interest to selectively propagate
  • Give colonies room to grow in reprogramming experiments by removing adjacent fibroblasts
  • Increase the number of iPS cell lines that be generated in an experiment
  • Clone specific colonies when generating genetically modified stem cell line
  • In situ laser processing is done with the microplate lid in place, reducing the risk of contamination
